Not only can literally every one of us know this by tasting the wine (or grape juice, as the case may be), but if it were literally true that wine became blood and bread became flesh, that would be cannibalism! Throughout the Old Testament, cannibalism is represented as a dreadful curse, an awful consequence received by rebellious jews at their worst, and this is highly incongruous with Jesus' message in the Lord's supper, which is one of a blessing.
